C-Peptide  Sample Requirements & Methods 

 

Sample Preparation 

  • ♦ Serum – Use a serum separator tube (SST).  Let samples clot 30 minutes before centrifugation at 1000 × g for 15 minutes. Remove serum immediately, aliquot, and store samples at  -80°C. Avoid freeze-thaw, the assay should be run on the first thaw.
  • ♦ Plasma – Use EDTA or heparin as an anticoagulant to collect plasma (grey top vacutainer). Within 30 minutes of drawing sample, centrifuge for 15 minutes at 1000 × g at 2 – 8°C. Remove plasma immediately and store samples at  -80°C. Avoid freeze-thaw, the assay should be run on the first thaw.
  • ♦ Each assay requires different volumes of plasma or serum, please see table below. The volume listed allows for repeat measures if necessary.
  • ♦ Label samples with your identification number and the time and date that it was drawn. If you are measuring area under the curve, the exact time samples were drawn (not 90 mins for example, but 8:35 AM) should be on label and if possible, in a digital file.

Note: Grossly hemolyzed samples cannot be measured.

 Methods of Analysis

Mitokine uses commercially available assays for C-peptide as described in the table below. Assays are performed according to the manufacturers’ recommendations.

Quality Control 

  • ♦ All standards, controls and samples are run in duplicate. The Coefficient of Variance (CV) is calculated for each set of duplicates. The CV must meet the laboratory’s Quality Control criteria established for the assay or the sample will be re-analyzed (if sufficient volume is available). The average C-peptide concentration of the duplicates is reported to the Principal Investigator.
  • ♦ Manufacturer controls are included in each assay along with our own internal laboratory controls.

Packing Samples

  • ♦ Pack on plenty of dry ice and seal the cooler lid with packing tape so the specimens remain frozen should the shipment be delayed.
  • ♦ Ship overnight no later than Wednesday, the lab is closed on weekends
  • ♦ Pack specimens in sample boxes, plastic bags often break in contact with dry ice.
  • ♦ Include and email quantify@mitokine.com a copy of the sample inventory. This will help us confirm that all shipments are complete.
  • ♦ Please email quantify@mitokine.com or phone (207) 422-6838 when you ship so that someone is available to receive samples.
